Ambrisentan and tadalafil synergistically relax endothelin-induced contraction of rat pulmonary arteries.

Liang F, Yang S, Yao L et al.
In this experimental study, using male Sprague–Dawley rats, the researchers tested two hypotheses: firstly, that the combination of ambrisentan and the phosphodiesterase-5 inhibitor tadalafil may be more effective than either drug alone to reduce an ET-1-induced increase in pulmonary vascular constriction; and secondly, that the synergistic effects of these drugs depend on the presence of both ETA and ETB receptors.
